What would it be?
well lets make it a little easier...
one revolver
one semi-auto
And why would you chose that particular gun?
if you could only own ONE handgun...

A Webley chambered in .455 and one of those (Nationalist) Chinese Broomhandles chambered in .45 . Full auto in a small package.

...that would suck.if you could only own ONE handgun...
If someone in any future that I am still a part of tries to make that one of the rules, I will not follow.
As for most favored auto it would be one of the many well made 1911s in .45acp...just as JMB (pbuh) designed it.
Revolver would be a Ruger GP100 - reliable as a sledge hammer and sorta built like one too.
Re: if you could only own ONE handgun...
For my semi-auto, I would take a SIG P-210- especally one of the ones with the US styled mag releases and heavy frame.
For a revolver, a Colt Python 6".
Both are accurate, well made, and fine examples of their respective types.

Re: if you could only own ONE handgun...
I'm not so fond of these "only one (more)" threads. It's kinda like asking if you could only have one hammer. I'd feel pretty silly trying to peen over a rivet with a claw hammer, or trying to drive a tack with a sledge hammer.
That being said, I'm going to have to agree with CBryneIV on a Colt 1911 and/or an older model S&W 686.
Because quality equals reliability, and either can be used for various purposes. The ammo is cheap which means more target practice, and can be concealed with only minor difficulties.
Neither can do everything, and thats the point of having a large "toolbox."

You didn't say whether ammo availability was a concern/criterion.
Going on the assumption that "ammo fairies" would supply me with all the ammo I could need:
* revolver -- Freedom Arms 83, 8", 454 Casull -- biggest cartridge I can handle easily; defense, hunting, etc.
* semi-auto -- 45 Super, or 10mm 1911 platform.

Ruger Redhawk 4 inch stainless 44 mag, 44 special. Since the Redhawk could handle defense and " Big Game " , I'll go with a 22 semi auto, Ruger MK whatever with 6 inch bull barrel, for fun and little stuff.

One and only handgun: Smith and Wesson .44 Magnum or .45 Colt N frame, Something like the Mountain gun perhaps.
One Revolver One Auto in a world where concealed carry is still "allowed":
Revolver: S&W airweight bodyguard (shrouded hammer)
Auto: 1911
One Revolver One Auto in a world where concealed carry is not "allowed":
Revolver: Freedom Arms Model 83 in .475

My Rohrbaugh. If I'm going to only have one, I want one that I can carry wearing anything.
Revolver... SP101 3"
One auto and one revolver -- Ruger Redhawk in 45 Colt and a 1911 in 45 ACP.
